More on W-18's pharmacology

So, no predicted opioid activities again. Only histamine H3 receptor activity. And data will be posted on Bioarchivx sometime this coming week!

  2. How about nicotinic acetylcholine receptors as causes of or contributors to analgesic effects and toxicity? (Recall the non-opioid epibatidine active in the (non-specific) Straub tail test.)
    I have (intentionally?) stumbled upon a recent paper describing structure - insecticidal activity relationship studies on very close relatives of the W-x compounds.

  3. The W-18 is probably devoid of opioid activity. The "data" came from Canadian university group publication of their test results in mouse writhing and hot plate tests. It was never re-confirmed, and now it turns out to be just an artifact.

    These mouse pain perception tests can be easily skewed by any off-target central activity that makes the mouse incapacitated. In this case, histaminic activity-related drowsines
